I've never seen Izzard dance. I've seen a couple of his stand-up shows, and pretty much all I've seen him do is ... stand up. In a dress. (Actually I think I've mostly seen him wear a skirt.) I mean, he's funny and all, but would he really have the chops to play the part?
But beyond the question of who could pull a particular role off, while a new stage revival is certainly workable, my main issue with this is that as a film, the quintessential version has already been done. A remake will never capture what the film accomplished. A great deal of its charm is the dated cheesiness.

Sock - See Revengers Tragedy or Cat's Meow or his series The Riches on FX, but especially Revengers Tragedy. He's a talented actor and getting better the more he works at it. While I am confident he could pull the role off, I seriously doubt he would do it. Which means I won't be forced to support this travesty, which is fine with me.
Legs - I've never actually thought of that. I prefer Stewie's cookie whore girlfriend anyway, possibly my favorite fictional Janet of all.
